چکیده

Background It is well known that in the yeast S. cerevisiae the sugars sucrose and maltose/maltotriose are metabolized by different pathways: sucrose is hydrolyzed by extracellular invertase (encoded by SUC genes), while maltose and maltotriose are actively transported into the cell and hydrolyzed by intracellular a-glucosidases (both proteins encoded by the MAL genes). Nevertheless, several reports have shown that some SUC genes can be located proximal to MAL genes at the telomeres of different chromosomes. Furthermore, the SUC genes also allow the synthesis of an intracellular form of invertase, an enzyme with no obvious function in yeasts [1]. We have already shown that sucrose can be metabolized by yeast cells through MAL-encoded transporters and a-glucosidases.

Molecular analysis of maltotriose active transport and fermentation by Saccharomyces cerevisiae reveals a determinant role for the AGT1 permease.

Incomplete and/or sluggish maltotriose fermentation causes both quality and economic problems in the ale-brewing industry. Although it has been proposed previously that the sugar uptake must be responsible for these undesirable phenotypes, there have been conflicting reports on whether all the known alpha-glucoside transporters in Saccharomyces cerevisiae (MALx1, AGT1, and MPH2 and MPH3 transpo...

EXPRESSION OF Saccharomyces cerevisiae α-GLUCOSIDE TRANSPORTERS UNDER DIFFERENT GROWTH CONDITIONS

Important biotechnological processes depend on the efficient fermentation by Saccharomyces cerevisiae yeasts of starch hydrolysates rich in maltose and maltotriose. The rate-limiting step for fermentation of these α-glucosides is the transport across the plasma membrane of the cells. In order to contribute to a better understanding of maltose and maltotriose metabolism by S. cerevisiae, we anal...

Switching the mode of sucrose utilization by Saccharomyces cerevisiae

BACKGROUND Overflow metabolism is an undesirable characteristic of aerobic cultures of Saccharomyces cerevisiae during biomass-directed processes. It results from elevated sugar consumption rates that cause a high substrate conversion to ethanol and other bi-products, severely affecting cell physiology, bioprocess performance, and biomass yields. Fed-batch culture, where sucrose consumption rat...

Invertase Production from a Hyperproducing Saccharomyces Cerevisiae Strain Isolated from Dates

Invertase production by wild cultures of Saccharomyces cerevisiae isolated from dates available in local market is reported. Five hyperproducing yeast strains (>100 fold higher invertase activity) were kinetically analyzed for invertase production. Saccharomyces cerevisiae strain GCAII was found to be a better invertase yielding strain among all other isolates. The values of Qp and Yp/s for GCA...

Molecular analysis of maltotriose transport and utilization by Saccharomyces cerevisiae.

Efficient fermentation of maltotriose is a desired property of Saccharomyces cerevisiae for brewing. In a standard wort, maltotriose is the second most abundant sugar, and slower uptake leads to residual maltotriose in the finished product. The limiting factor of sugar metabolism is its transport, and there are conflicting reports on whether a specific maltotriose permease exists or whether the...
